A compound that switches the bone-building pathway on in two ways, one by blocking GSK3 and one at a brake that builds up as a plate ages.
Small molecule that raises Wnt/β-catenin in two steps: it inhibits GSK3β, which lifts the destruction complex directly, and then interferes with the CXXC5–DVL interaction, removing the brake that accumulates as a plate ages.
Risks. Nothing is known about its safety in people; in the mice, joint cartilage and liver were undamaged.
