Wnt and β-catenin

The switch that converts cartilage into bone, the LRP5 and LRP6 co-receptors behind it, the contested claim that LRP5 acts on bone from the gut, why forcing it shortens a bone while lifting a brake lengthens one, and the drugs built for density against the one aimed at length.

Wnt and β-catenin: without a signal, sclerostin and DKK1 block LRP5/6 and a destruction complex of GSK3, APC and Axin destroys β-catenin. When Wnt binds Frizzled and LRP5/6, β-catenin builds up and reaches the nucleus. sclerostin, DKK1GSK3, APC, AxinWntTCF/LEFOffOnβ-catenin destroyedβ-catenin reaches the nucleus

Wnt signalling decides that a cell builds bone. It turns the cartilage template into the mineralised shaft, and osteoporosis drug development has spent two decades learning to push it. For height, it governs the conversion of cartilage to bone rather than the supply of new cartilage.
