A laboratory chemical found by screening, which switches on cartilage genes and holds down the ones that turn cartilage into bone.
A thienoindazole found in a screen of roughly 2,500 compounds against a collagen II reporter. It induces Runx1, which binds the COL2A1 promoter and raises cartilage matrix genes while holding down Col10a1, Alp and Mmp13. A research compound with no registered trial.
Risks. No safety data at all. Its effect flips direction across a narrow range, and the gene it acts on is one involved in leukaemia.
