RUNX2, MEF2C and HDAC4

The transcription factors that commit a chondrocyte to becoming bone, the deacetylase that holds them off, SP7 downstream of RUNX2, the remodeller CHD9 that sets how much RUNX2 is made, and what happens when that restraint is removed.

RUNX2, MEF2C and HDAC4: HDAC4 binds RUNX2 and MEF2C and holds them down. Released, they drive hypertrophy and the switch from cartilage to bone. HDAC4RUNX2MEF2CRUNX2MEF2CHeldReleasedCartilage to boneHDAC4 binds bothbone genes onhypertrophy first

By the time a chondrocyte is ready to hypertrophy, the decision is being made by transcription factors rather than by signals arriving from outside. RUNX2 is the master regulator of the osteoblast programme and the driver of terminal chondrocyte maturation. MEF2C works alongside it. HDAC4 stops them firing early.
