Pioglitazone / Rosiglitazone
Approved, for something elseMeasured in people, not heightBy mouthSmall molecule
Two diabetes tablets that switch the fat-or-bone factor on. Licensed for blood sugar in adults, and for nothing skeletal.
Thiazolidinediones, sold as Actos and Avandia: oral PPAR-γ agonists, approved as an adjunct to diet and exercise to improve glycaemic control in adults with type 2 diabetes. Neither is licensed for anything skeletal.
Risks. Both carry the strongest class of warning there is, for heart failure, and neither is to be started once heart failure is advanced. Rosiglitazone's label counts 9.3% of women breaking a bone over four to six years against 3.5% on an older diabetes tablet. Pioglitazone is not recommended for children at all, on the strength of what it does to adults, and rosiglitazone's label says the data are too thin to recommend it for children either.
