A vitamin A-like drug built to hit one receptor subtype, approved for a rare disease in which bone forms inside soft tissue.
An orally bioavailable retinoic acid receptor agonist with particular selectivity for the gamma subtype. Through RARγ it decreases BMP/ALK2 signalling by inhibiting SMAD1/5/8 phosphorylation, reducing the chondrogenesis that produces heterotopic bone. Approved for reduction in the volume of new heterotopic ossification in fibrodysplasia ossificans progressiva, in adults and in children aged 8 and over for females and 10 and over for males; first approval was Canada, 21 January 2022.
Risks. It carries the strongest kind of label warning, for harm to a fetus and for closing a growing child's growth plates early, with x-rays and height checks required before and during treatment.
