MCC950 and TAK-242

Not approved for people anywhereMeasured in people, not heightResearch onlySmall molecule
Two laboratory compounds that block the alarm systems which switch inflammation on, rather than blocking the inflammatory signals themselves.
Two compounds that block the machinery detecting danger rather than the cytokines themselves. MCC950 inhibits the NLRP3 inflammasome, the complex that activates caspase-1 and so releases mature IL-1β. TAK-242 blocks TLR4, the receptor that detects bacterial products and fragments of broken-down matrix.
Risks. Neither is approved. TAK-242 was tried in patients with sepsis, where it raised a form of haemoglobin that cannot carry oxygen, and switching off these alarms also switches off part of the defence against infection.
