A laboratory chemical that fills the slot BRD4 uses to grip acetyl marks, so the reader comes off the DNA. Not a medicine anywhere.
A thienotriazolodiazepine that competes for the acetyl-lysine binding pocket of the BET bromodomains, displacing BRD4 from acetylated histones. A research reagent, never a drug: approved nowhere, and no registered trial has used it.
Risks. Nobody has taken it and it is approved nowhere, so there is no label and no boxed warning. In mice it stops bone cells maturing on both sides of bone turnover, and losing the protein it blocks thins the growth plate. In male mice it shrank the testes and cut sperm numbers, reversing after it was stopped.
