Histone acetylation and methylation at the growth plate, DNA demethylation, BET readers, and the nucleosome remodelling faults that shorten a skeleton.

DNA is wound round histone spools, and SOX9 cannot reach a stretch a spool covers. CBP and p300 hang acetyl marks on the spool and the HDACs take them off; BRD4 reads a mark, BAF slides the spool aside, and SOX9 reaches COL2A1. The nucleosome is the unit: 147 base pairs of DNA wound round a histone octamer. What a transcription factor can reach is set by where those octamers sit and by the marks already carried on them and on the DNA underneath. The marks on RNA rather than DNA are in METTL3 and m6A, which also holds the chromatin overgrowth syndromes, and HDAC4's hold on RUNX2 is in RUNX2, MEF2C and HDAC4.
