Erdafitinib / Pemigatinib / Futibatinib

Approved, for something elseMeasured in people, not heightBy mouthSmall molecule
Three approved cancer drugs that switch off a whole family of growth-signal receptors at once.
Three pan-FGFR kinase inhibitors, all of them approved cancer drugs. They inhibit multiple FGFR isoforms, preventing receptor trans-autophosphorylation. Erdafitinib and pemigatinib bind reversibly; futibatinib covalently and irreversibly.
Risks. They are cancer drugs and they do cancer-drug damage: fluid under the retina in a fifth of patients, phosphate high enough in three quarters of them to risk calcium depositing in soft tissue, and nail damage in seven out of ten.
