An old epilepsy drug that happens to make cells dispose of faulty proteins faster, which is the only reason it was tried for a cartilage disease.
A long-established anticonvulsant that stimulates intracellular proteolysis, which is the whole reason it was repurposed for a collagen disease. Approved for partial seizures with complex symptomatology, generalised tonic-clonic seizures and mixed seizure patterns, and for the pain of true trigeminal neuralgia; the extended-release capsule is additionally approved for acute manic or mixed episodes associated with bipolar I disorder. There is no approval for any skeletal indication, though the European Medicines Agency granted orphan designation for it in metaphyseal chondrodysplasia type Schmid.
Risks. The label carries the strongest warnings for life-threatening skin reactions in people with a particular genetic variant, and for bone marrow failure.
