AXL and BGB324
The receptor that holds osteoblasts back from maturing, the Isg15 and Erk axis below it, and what blocking it with BGB324 did to bone.
A receptor tyrosine kinase of the TAM family — Tyro3, Axl, Mer — activated by its ligand Gas6 and signalling through PI3K/AKT and MAPK. It is well studied in cancer, immune regulation and vascular biology, and its role in bone went largely unexamined until an unbiased RNAi screen across the mouse kinome picked it out as a negative regulator of osteoblast differentiation.
